Overview

Count cells manually at a fraction of the cost and footprint of automated cell counters by using the Hausser Scientific™ Bright-Line Hemocytometer. With one-piece construction to ensure long-term durability, the metallized chamber enables accurate determination of cell concentrations for culture plating density, determination of cell viability, and assessment of cell isolation procedure results. This hemocytometer has an H-shaped moat that forms two counting areas, or plateaus. Each plateau features enhanced Neubauer rulings and a “V” slash at the loading side that facilitates sample loading and reduces the risk of overflow into the moat. Contact of the flat, polished cover glass surfaces with cover glass supports produces an exact volume of fluid over the counting area. A ruled surface 0.1 mm below the cover glass limits the volume of blood or fluid over a square mm at 0.1 cubic mm; the volume over the 400 squares in each counting area is 0.00025 cubic mm. The difference in surface tension characteristics between the cover glasses and the metallic surface of the chamber ensures smooth capillarity for precise loading and even cell distribution.

Cell counting can be performed using Trypan Blue (for total viable mammalian cell count; Catalog #07050) or 3% Acetic Acid with Methylene Blue (for total nucleated cell count; Catalog #07060). Cell recovery can be calculated based on cell counts before and after isolation. The viability of cells should be evaluated when a potential decrease in viability may be expected, such as when working with cryopreserved cells or ex vivo-manipulated cells.
