Overview

The STEMdiff™ Astrocyte Maturation Kit is used in conjunction with the STEMdiff™ Astrocyte Differentiation Kit (Catalog #08540) to rapidly and efficiently generate astrocytes from neural progenitor cells derived from human pluripotent stem cells using the STEMdiff™ Neural Induction Medium (Catalog #05835) embryoid body protocol. Using this system, a highly pure population of astrocytes (>85% GFAP-positive astrocytes; <15% class III β-tubulin-positive neurons) can be generated from hPSCs in as few as 7 weeks and can be maintained long-term in culture. Cells derived using these products are versatile tools for modeling human neurological development and disease, drug screening, toxicity testing, and cell therapy validation.

Figure 1. Downstream Differentiation of Neural Progenitor Cells to Astrocytes

Neural Progenitor Cells derived from ES cells (H9 cell line) using the STEMdiff™ Neural Induction Medium embryoid body protocol were differentiated and matured to astrocytes with typical star-like morphology by culture in STEMdiff™ Astrocyte Differentiation Kit for 19-20 days and STEMdiff™ Astrocyte Maturation Kit for 56 days. The astrocytes express GFAP (green) but not the neuronal marker class III β-tubulin (red). Nuclei are counterstained with DAPI.
