Overview

The principal physiological regulator of erythropoiesis, Erythropoietin (EPO) is a glycoprotein growth factor that is produced primarily in the kidney in response to hypoxia or anemia. EPO promotes erythropoiesis by binding to a homodimeric cell surface receptor that activates JAK2/STAT5, PI3K/AKT, and MAPK pathways, and stimulates the proliferation and differentiation of erythroid progenitor cells (Jelkmann. Transfus Med Hemother, 2013; Kuhrt & Wojchowski. Blood, 2015).

(A) The biological activity of Human Recombinant EPO was tested by its ability to promote the proliferation of TF-1 cells. Cell proliferation was measured using a fluorometric assay method. The EC50 is defined as the effective concentration of the growth factor at which the cell proliferation is at 50% of maximum. The EC50 in the above example is less than 0.6 ng/mL. (B) Human Recombinant EPO stimulates the proliferation and differentiation of erythroid progenitor cells, and was validated by titration in a CFU assay on human bone marrow MNCs, using MethoCult™ SF H4236 (Catalog #04236). The total number of colonies at each EPO concentration was enumerated using STEMvision™ (Catalog #22006). (C) 5 μg of Human Recombinant EPO was resolved with SDS-PAGE under reducing (+) and non-reducing (-) conditions and visualized by Coomassie Blue staining. Human Recombinant EPO polypeptide has a predicted molecular mass of 18.4 kDa. As a result of glycosylation, the recombinant protein migrates with an apparent molecular mass of 26 - 36 kDa in SDS-PAGE.
