Start experiments when you want to with large numbers of leukocytes, including peripheral blood mononuclear cells (PBMCs). As a cryopreserved product, frozen leukopaks provide samples from specific donors that can be processed when the end-user is ready. Leukopaks are available in Full (≥ 9 billion cells), Half (≥ 5 billion cells), Quarter (≥ 2.5 billion cells), and Tenth (≥ 900 million cells) size formats. Each frozen leukopak is a highly concentrated, low-volume apheresis collection from a single, normal donor, using Institutional Review Board (IRB)-approved consent forms and protocols. Each full-sized leukopak is produced from ~2 - 3x blood volumes using the Spectra Optia® Apheresis System, with acid-citrate-dextrose solution A (ACDA) as the anticoagulant. The collected product is then cryopreserved in a controlled-rate freezer. High-resolution HLA typing (Class I and Class II alleles and CMV status) are available upon request. Donor specifications (e.g. BMI category, smoking status, ethnicity, etc.) can be requested in the comment box above, after selecting from the product options. Donors are screened for HIV-1, HIV-2, hepatitis B, and hepatitis C.

What is the percentage of viable cells in a frozen leukopak?
Upon cryopreservation, cell viability in the frozen leukopak is greater than 90%. After thawing and the cleanup process, the viability is usually greater than 90%.
How does a frozen leukopak differ from a fresh leukopak?
Fresh leukopaks are shipped immediately after collection for same-day processing, while frozen leukopaks are cryopreserved and stored in vapor-phase liquid nitrogen until use. Frozen leukopaks allow for flexible experimental timing, batch testing across multiple donors, and long-term inventory planning. Fresh leukopaks may be preferred when immediate processing and maximal functional activity are required. Learn more about leukopaks here.
Are there any advantages of working with frozen leukopaks compared to fresh leukopaks?
Both fresh and frozen leukopaks are ideal starting materials for downstream cell isolation when large numbers of cells are required, reducing the time and reagents needed to process cells of interest. However, frozen leukopaks provide additional convenience by allowing you to start experiments according to your schedule. Additionally, cell viability may be better preserved in frozen leukopaks during global shipping. Frozen leukopaks may also provide you with access to samples from the same lot at a future date.
How should frozen leukopaks be thawed and handled?
Frozen leukopaks should be rapidly thawed using validated protocols to maximize post-thaw recovery and viability. Following thaw, PBMCs can be isolated or immune subsets enriched using immunomagnetic separation methods. Proper thawing and handling are critical to preserve cell functionality for downstream immune assays and expansion workflows.
What are frozen leukopaks used for?
Frozen leukopaks are commonly used in: CAR-T and CAR-NK research; T cell expansion and activation; NK cell cytotoxicity and potency assays; ADCC assays; gene editing and transduction workflows; and assay standardization across donor cohorts. They are particularly useful when consistent donor material is required over time.
What donor screening and documentation are available for frozen leukopaks?
Frozen leukopaks are sourced from consented healthy donors under IRB-approved protocols. Donor eligibility criteria, demographic information, and infectious disease screening documentation are available to support research traceability and compliance.
