Using Cell Density Gradients to Optimize Your Cell Isolations

RosetteSep™ is a unique immunodensity cell isolation platform that isolates highly purified cells during density gradient centrifugation. With RosetteSep™, isolating untouched cells from human whole blood samples is fast, easy, and uses no columns or magnets.

In order to optimize your cell isolations using RosetteSep™, we recommend using the appropriate density medium. There are a number of different density media available to suit your particular needs:

Name of Gradient Medium

Catalog #

Density

Cells Isolated

Lymphoprep™ *

07801 07811 07851 07861

1.077 g/mL

Recommended for the isolation of mononuclear cells (MNCs) from peripheral blood, cord blood, and bone marrow by exploiting differences in cell density. Also suitable for the enrichment of a number of MNC subtypes using RosetteSep™.

Ficoll-Paque™

1.077 g/mL

Recommended for the isolation of mononuclear cells (MNCs) from peripheral blood, cord blood, and bone marrow by exploiting differences in cell density. Also suitable for the enrichment of a number of MNC subtypes using RosetteSep™.

RosetteSep™ DM-L Density Medium

15705

1.081 g/mL

RosetteSep™ DM-L is a density gradient medium designed specifically for the enrichment of human lymphocyte populations (CD3+ T cells, CD4+ T cells, CD8+ T cells, and B cells) from whole blood using the following with RosetteSep™ cocktails:

RosetteSep™ DM-M Density Medium

15725

1.085 g/mL

RosetteSep™ DM-M is a density gradient medium designed specifically for the enrichment of human myeloid (CD33+) cells from whole blood using the following RosetteSep™ cocktail:

* Lymphoprep™ is a trademark of Axis-Shield. Lymphoprep™ is equivalent to Ficoll-Paque™ in composition and has the exact same density. Lymphoprep™ can be used interchangeably with Ficoll-Paque™ for RosetteSep™ separations.

These density media can all be used in conjunction with our SepMate™ tubes, for quick and easier layering of blood, allowing for consistent and hassle-free density-based cell separation in as little as 15 minutes.
