Mammalian Cell Cloning Procedure: Customizable Semi-Solid Cloning with ClonaCell™ FLEX
Cloning mammalian cells using semi-solid cell culture media can be an efficient way of generating monoclonal cell lines for biopharmaceutical development. Semi-solid cloning physically separates individual cells by suspending them in the viscous medium, allowing them to grow into discrete colonies that can be easily isolated. This method enables isolation of a large number of diverse clones, each with a high probability of monoclonality, in less time and using fewer resources than limiting dilution cloning. This video outlines the ClonaCell™ FLEX procedure for cloning in a customized semi-solid cloning medium that is based on your own liquid cell culture medium formulation. Steps covered in the video include: preparation of the custom semi-solid medium, preparation of the cells, plating the semi-solid cell suspension, colony picking and expansion prior to screening for the desired monoclonal antibody or other protein product of interest.
