Overview

Phalloidin is a heptapeptide derived from the poisonous mushroom Amanita phalloides. Phalloidin binds with a high specificity to F-actin filaments in cells, preventing filament depolymerization. Fluorescently labeled phalloidin can be used in labeling, identification, and quantitation of F-actin in formaldehyde-fixed and permeabilized cells. Fluorescently labeled phalloidin has lower non-specific binding compared to actin antibodies across most species of plants and animals, which provides low background and high contrast during imaging.

H9 embryonic stem cells were plated in mTeSR™ Plus (Catalog #100-0276) on Corning® Matrigel®-coated 35 mm glass-bottom dishes for 24 hours, then fixed with 4% paraformaldehyde and stained with Phalloidin-iFluor™ 647 (red), and counterstained with Hoechst 33342 (blue).

WLS-1C cell-derived neural progenitor cells were differentiated to forebrain-type neurons using STEMdiff™ Forebrain Neuron Differentiation (Catalog #08600) and STEMdiff™ Forebrain Neuron Maturation Kits (Catalog #08605). Neurons were cultured on poly-L-ornithine/laminin-coated 35 mm glass-bottom dishes for 3 weeks, then fixed with 4% paraformaldehyde and stained with Phalloidin-iFluor™ 647 (red), and counterstained with Hoechst 33342 (blue).
