How to Assemble and Purge the Hypoxia Incubator Chamber

Provides a step-by-step procedure on how to assemble and purge the hypoxia chamber incubator & how to assess the resulting low-oxygen environment

At 3 minutes and 50 seconds of the video, it should state to adjust the regulator valve control to 20 L/min by using the flow meter and not to “~20 psi". This instructional video provides a step-by-step procedure on how to assemble and purge the Hypoxia Incubator Chamber and how to assess the resulting low-oxygen environment. A hypoxic environment mimics physiological conditions and many cell types have been shown to have enhanced proliferation when cultured with lowered atmospheric oxygen tension. For example, mouse mesenchymal stromal cells (MSCs) have been shown to be highly sensitive to atmospheric oxygen tension, hence culturing these cells under hypoxic conditions facilitates optimal clonogenicity and cell proliferation.
