Overview

Learn essential techniques and best practices for processing human samples to isolate high-quality, functional immune cell types for your downstream applications. This three-day, part-time, and live-streamed training is led by experts from STEMCELL Technologies and consists of interactive lectures and laboratory demonstrations. You’ll learn how to handle, isolate, analyze and cryopreserve your human immune cells quickly and efficiently. You will also have the opportunity to interact with trainers and fellow participants throughout the sessions.

Learning Outcomes:
• Become familiar with commonly available human blood products
• Understand how different human blood products are generated and how they differ in cellular composition
• Be aware of standard methods for processing blood products to obtain leukocytes for downstream applications
• Understand how blood sample processing methods influence the resulting leukocyte populations
• Compare between different technologies that can be used to isolate specific human immune cell subsets
• Be aware of the different parameters that may influence the choice of the cell isolation strategy
• Understand the theory behind common downstream assays of isolated immune cells and their applications
• Gain a basic understanding of flow cytometry and how it is applied to immune cell analysis
• Apply best practices for setting up the proper controls when using flow cytometry to assess isolated cells

Day 1 (2.5 hrs, including introduction, break, and Q&A)

  • Lecture: Introduction to commonly sourced human blood products and how they compare (0.5 hrs)
  • Lecture: Overview of common processing methods used for human blood products (0.5 hrs)
  • Virtual Lab: Preparation of human peripheral blood mononuclear cells (PBMCs) by density gradient separation (1 hr)

Day 2 (4.25 hrs, including break and Q&A)

  • Lecture: Overview of cell isolation technologies and best practices (1 hr)
  • Virtual Lab: Preparation of human PBMCs directly from whole blood using immunomagnetic cell isolation technology (0.5 hrs)
  • Virtual Lab: Cryopreservation of human PBMCs (0.5 hrs)
  • Virtual Lab: Thawing of human PBMCs for downstream applications (0.5 hrs)
  • Virtual Lab: Isolation of human T cells from PBMCs (0.5 hrs)

Day 3 (4.25 hrs, including break and wrap-up, and Q&A)

  • Lecture: Downstream applications of isolated immune cells (1 hr)
  • Lecture: Introduction to flow cytometry theory and best practices assessing isolated cells (1 hr)
  • Virtual Lab: How to culture and expand human T cells in serum-free medium and parameters used to assess T cell activation and expansion (1 hr)

This course is certified by the United Kingdom CPD Certification Service, which may be applicable to individuals who are members of, or are associated with, UK-based professional bodies. This course consists of approximately 8 hours of learning.
