Monitor superoxide production in the mitochondria of live cells using Mitochondrial Superoxide Dye. This fluorescent dye is a valuable tool for detecting mitochondrial superoxide production to investigate oxidative stress, and has been verified for detection using flow cytometry. Reactive oxygen species (ROS) and reactive nitrogen species (RNS) are free radicals derived from oxygen and nitrogen molecules.

Mitochondrial Superoxide Dye selectively targets the mitochondria of living cells. This dye becomes oxidized when it reacts with superoxide, a ROS, producing a red fluorescence. In the absence of superoxide, there is no absorption and negligible fluorescence. Though Mitochondrial Superoxide Dye is readily oxidized by superoxide, it is resistant to oxidation by other ROS and RNS. The high signal-to-noise ratio makes Mitochondrial Superoxide Dye a suitable reagent for superoxide detection in biological systems such as cellular respiration (Li et al. Front Immunol, 2018).
