The RosetteSep™ and SepMate™ system is an innovative platform for rapid and consistent cell isolation directly from whole blood in just 25 minutes. The system combines a unique immunodensity cell isolation reagent (RosetteSep™) with a specialized cell processing tube (SepMate™) to minimize variability between users and allow efficient, high-throughput sample processing. This new method isolates untouched and highly purified cells in a single step without columns or magnets, helping to avoid the risk of activating or damaging cells.

Cells isolated using the RosetteSep™ and SepMate™ system are immediately available for downstream use, and are ideal for cytotoxicity assays, compound screening, and other high-throughput applications that require viable, functional cells with minimal manipulation.

How RosetteSep™ Works

To isolate immune cells from whole blood, simply incubate your sample with the appropriate RosetteSep™ antibody cocktail prior to SepMate™ separation or standard density gradient centrifugation. RosetteSep™ crosslinks unwanted cells to red blood cells, forming immunorosettes (Figure 1). These immunorosettes pellet during centrifugation, leaving untouched, highly purified target cells at the interface between the plasma and the density gradient medium. If you are using SepMate™, simply pour purified cells into a new tube. Otherwise, harvest them from the plasma:density gradient medium interface.

The Novel RosetteSep™ and SepMate™ Procedure


Incubate your blood samples with RosetteSep™ for 10 minutes. Rapidly load samples into SepMate™ tubes. The unique SepMate™ insert prevents the blood from mixing with the density gradient medium. After a 10-minute centrifugation with the brake on, simply pour highly purified target cells into a new tube.
