Toxicity Testing in Hematopoietic Progenitors with Colony Assays (CFC Assays)

An important goal during pre-clinical drug development is to predict whether a new agent will be clinically toxic to the bone marrow and whether the toxicity will be specific to one cell lineage (lymphocytes, neutrophils, megakaryocytes or erythrocytes). Other important goals are to determine at what dose or plasma level the drug will be toxic, which model best predicts the clinical situation, and when onset of cytopenia and recovery will be likely to occur.

The CFU-GM assay can be used as a surrogate assay to test myelosuppressive agents and has been shown to accurately predict acute neutropenia in humans. The European Centre for the Validation of Alternative Methods (ECVAM) endorses the use of the colony-forming unit-granulocyte/macrophage (CFU-GM) assay for this application as an alternative for drug testing in animals.

Primary human cells

Primary human cells are a valuable tool for researchers and are often preferred over transformed or immortalized cell lines because they are more representative of cells in vivo. A wide range of frozen (unprocessed and purified) primary human cells is available, including primary hematopoietic cells from bone marrow (BM), peripheral blood (PB), and umbilical cord blood (CB). Marrow stromal cells are also available.

ClonaCell EasyPick – Automated Cell Line Generation

Traditional methods of selecting and cloning hybridomas and transfected cells are performed in liquid culture and require several limiting dilutions and screening steps. ClonaCell^® methylcellulose-based semi-solid media can combine selection and cloning in one easy step. Using ClonaCell^®, stable cell lines can be generated in as little as 14 days. Selection of clones can be fully automated using the ClonaCell^® EasyPick platform.
