Overview

TFF-Easy is a tangential flow filtration (TFF)-based EV concentrator that efficiently removes small molecules and buffer components while retaining extracellular vesicles (EVs) and other desired sample constituents. This concentrator is suitable for processing samples from various sources, including cell culture conditioned media, plasma, urine, and serum.

TFF-Easy can be used upstream of size-exclusion chromatography (SEC) to pre-concentrate samples and support downstream EV isolation. It also enables dialysis for complete buffer exchange, offering flexibility in sample preparation workflows. Concentrated samples can be easily recovered using a syringe. For small volumes, filtration can be performed manually on the benchtop, while larger volumes can be processed by connecting TFF-Easy to a peristaltic pump.

The product is supplied sterile and can be re-sterilized by beta irradiation if needed.

TFF-Easy and Centrifugal Filter, MWCO 100 kDa, Non-Sterile, 15 mL were used to concentrate particles from diluted plasma samples. Particle counts of the resulting concentrated samples were measured by microfluidic resistive pulse sensing (MRPS) and normalized to the input sample, which was set to 100%. Both TFF-Easy and Centrifugal Filter demonstrate high and comparable recovery of particles across two donors. Bars indicate mean relative particle count ± SD.

Spent medium from mesenchymal stromal cell (MSC) cultures was concentrated using either TFF-Easy or a Centrifugal Filter, MWCO 100 kDa, Non-Sterile, 15 mL, followed by extracellular vesicle (EV) isolation using the 2 mL Extracellular Vesicle SEC Column. EV-enriched SEC fractions were pooled prior to particle analysis. Particle counts of the pooled fractions were measured by microfluidic resistive pulse sensing (MRPS) and normalized to the centrifugal filter-concentrated sample, which was set to 100%. Both TFF-Easy and the Centrifugal Filter demonstrate comparable EV particle counts from two MSC donors following SEC isolation. Bars indicate mean relative particle count ± SD.

Spent medium from mesenchymal stromal cell (MSC) cultures was concentrated using either a Centrifugal Filter, MWCO 100 kDa, Non-Sterile, 15 mL, or TFF-Easy, followed by EV isolation using a 2 mL Extracellular Vesicle SEC Column. EV-enriched fractions were pooled and analyzed by Western blot under non-reducing conditions using the Extracellular Vesicle Human CD9/CD63/CD81 Antibody Panel, with albumin included as a non-EV protein control. Two loading volumes (15 µL and 30 µL) from the same pooled sample were analyzed for each concentration method. TFF-Easy and Centrifugal Filter demonstrate comparable capture of key EV markers from conditioned media
