Overview

CP-945,598 is a selective cannabinoid type 1 (CB1) receptor antagonist (Ki = 0.7 nM) and exhibits low affinity for CB2 receptors (Ki = 7.6 µM; Hadcock et al.). Cannabinoid receptors are part of the G protein-coupled receptor (GPCR) superfamily, and CB1 receptors are abundant in the brain and are able to attenuate neurotransmission (Mackie). Endocannabinoid system signaling components are expressed in the basal ganglia and interact with both GABA and dopaminergic signaling pathways. Modulation of CB1 is a potential therapeutic target for Parkinson’s disease (Han et al.).
