Overview

Centrifugal Filter, MWCO 100 kDa, Non-Sterile, provides a high sample recovery (> 90%), low protein binding, and chemical compatibility to a wide range of chemicals. Available in 4 mL and 15 mL sizes, these filters supply fast and efficient sample purification and concentration with a large membrane surface area consisting of two vertical polyethersulfone (PES) membranes.

For workflows requiring continuous processing of larger sample volumes, see our tangential flow filtration (TFF) (Cat #100-2460).

Data Figures

TFF 5-15 mL and Centrifugal Filter Recovery

Figure 1. TFF-Easy and Centrifugal Filter Achieve Comparable Particle Recovery

TFF-Easy and Centrifugal Filter, MWCO 100 kDa, Non-Sterile, 15 mL were used to concentrate particles from diluted plasma samples. Particle counts of the resulting concentrated samples were measured by microfluidic resistive pulse sensing (MRPS) and normalized to the input sample, which was set to 100%. Both TFF-Easy and Centrifugal Filter demonstrate high and comparable recovery of particles across two donors. Bars indicate mean relative particle count ± SD.

TFF 5-15 mL and Centrifugal Filter Relative Particle Counts

Figure 2. TFF-Easy and Centrifugal Filter Concentration of Conditioned Media Followed by SEC Isolation Yield Comparable EV Particle Counts

Spent medium from mesenchymal stromal cell (MSC) cultures was concentrated using either TFF-Easy or a Centrifugal Filter, MWCO 100 kDa, Non-Sterile, 15 mL, followed by extracellular vesicle (EV) isolation using the 2 mL Extracellular Vesicle SEC Column. EV-enriched SEC fractions were pooled prior to particle analysis. Particle counts of the pooled fractions were measured by microfluidic resistive pulse sensing (MRPS) and normalized to the centrifugal filter-concentrated sample, which was set to 100%. Both TFF-Easy and the Centrifugal Filter demonstrate comparable EV particle counts from two MSC donors following SEC isolation. Bars indicate mean relative particle count ± SD.

TFF 5-15 mL Western Blot

Figure 3. TFF-Easy and Centrifugal Filter Showing Comparable EV Marker Expression from MSC-Conditioned Media Following SEC Isolation

Spent medium from mesenchymal stromal cell (MSC) cultures was concentrated using either a Centrifugal Filter, MWCO 100 kDa, Non-Sterile, 15 mL, or TFF-Easy, followed by EV isolation using a 2 mL Extracellular Vesicle SEC Column. EV-enriched fractions were pooled and analyzed by Western blot under non-reducing conditions using the Extracellular Vesicle Human CD9/CD63/CD81 Antibody Panel, with albumin included as a non-EV protein control. Two loading volumes (15 µL and 30 µL) from the same pooled sample were analyzed for each concentration method. TFF-Easy and Centrifugal Filter demonstrate comparable capture of key EV markers from conditioned media
