Pure luminal organoid cultures (passage 1) were generated from non-sorted primary human mammary epithelial cells seeded in MammoCult™ Luminal Organoid Medium (LOM). The majority of organoids exhibit strong expression of luminal lineage markers MUC1, K8, ER, PR, and PMCA2 (n = 6 - 12), confirming their representative phenotype. The presence of secretory lineage luminal cells is validated by KIT expression (n = 10) and, more rarely, by co-expression of K8 and K14 (n = 3). These results demonstrate that MammoCult™ LOM enables the generation of physiologically relevant luminal mammary organoids that faithfully express lineage-specific markers, providing a powerful model for studying luminal cell biology and function.
