Monoclonal antibody production made easy! Since the first derivation of monoclonal antibodies of defined specificity in 1975, a variety of methods have been used to grow, select, and clone antibody-producing hybridomas. Many of the approaches are time-consuming and labor-intensive. Following fusion and growth in selective liquid medium, limiting dilutions are required to obtain monoclonal hybridomas. A problem often encountered is that some hybridomas overgrow others prior to cloning. Often, these faster growing cells do not synthesize antibodies, resulting in a failure to obtain the best antibody-producing hybridomas. A solution to these problems is to clone the hybridomas immediately after fusion using a selective semi-solid medium.
