NeuroCult® NS-A Proliferation Kit (Rat)
The NeuroCult® NS-A Proliferation Kit (Rat) is a standardized, serum-free medium for the culture of rat neural stem and progenitor cells as neurospheres. The NeuroCult® NS-A Proliferation Kit (Rat) must be supplemented with appropriate cytokines (not included). The medium is optimized to maintain rat neural stem cells in culture for extended periods of time without the loss of their self-renewal, proliferation or differentiation potential. The NeuroCult® NS-A Proliferation Kit (Rat) is comprised of NeuroCult® NS-A Basal Medium (Rat) and NeuroCult® NS-A Proliferation Supplement (Rat).
 
NOTE: Addition of EGF, Human, Recombinant (Catalog #02633), bFGF, Human, Recombinant (Catalog #02634) and Heparin (Catalog #07980) is required.

Cells cultured with the NeuroCult® NS-A Proliferation Kit (Rat) (NS-A Medium) continued to generate neurospheres beyond passage 15 resulting in an increase in total cell number. Cells cultured with a traditional formulation did not generate new neurospheres beyond passage 3 when cultured in T-25cm2 flasks.

Neurospheres could be continually generated in both media formulations beyond passage 10. However, higher cell expansion and percent viability were observed with the NeuroCult® NS-A Proliferation Kit (Rat) (NS-A Medium) compared to a traditional formulation. Neurospheres cultured in ultra-low adherent dishes were very difficult to dissociate after passage 4.


Rat NSCs cultured in NeuroCult® NS-A Proliferation Medium (Rat) maintain multi-lineage potential in long-term cultures and can be differentiated with NeuroCult® Differentiation Medium (Rat) to generate neurons, astrocytes and oligodendrocytes

Cells obtained from passage 10 neurospheres derived from E18 rat cortical cells cultured in NeuroCult® NS-A Proliferation Medium (Rat) were differentiated with NeuroCult® NS-A Differentiation Medium (Rat). Immunofluorescent staining was performed to identify the differentiated cell types.Neurons (red) were detected with a mouse monoclonal ß-Tubulin III antibody (Catalog #01409), oligodendrocytes (green) with a mouse monoclonal O4 Oligodendrocyte Marker antibody (Catalog #01416) and astrocytes (blue) with a rabbit polyclonal GFAP antibody (Catalog #01415).
