EasySep® box
The EasySep® HLA CD2 Positive Selection Kit is designed to isolate CD2+ cells from fresh or previously frozen human peripheral blood mononuclear cells by positive selection. Desired cells are targeted with Tetrameric Antibody Complexes recognizing CD2 and dextran-coated magnetic particles. The cocktail also contains an antibody to human Fc receptor to minimize nonspecific binding. Labeled cells are separated using an EasySep® magnet without the use of columns. Cells of interest remain in the tube while unwanted cells are poured off. The CD2 antigen is expressed on T cells and NK cells. Positively selected cells are compatible with flow cytometric crossmatch analysis (when used with EasySep® HLA FCXM Blocking Solution) and any other downstream assay.

  • Q. HOW DOES THE SEPARATION WORK?

    A. Tiny magnetic particles are attached to cells. When placed in the EasySep® Magnet, the cells migrate to the wall of the tube.

  • Q. HOW DOES THE BINDING OF THE NANOPARTICLE AFFECT THE FUNCTION OF CELLS? IS THE FUNCTION OF POSITIVELY SELECTED CELLS ALTERED BY THE BOUND PARTICLES?

    A. No, we have found that cells are not functionally altered.

  • Q. CAN EASYSEP® SEPARATIONS BE AUTOMATED?

  • Q. IN POSITIVE SELECTION CAN I DO MORE THAN 3 SEPARATIONS TO INCREASE PURITY?

    A. Yes, purity of targeted cells will increase with additional rounds of separations, however the recovery of targeted cells may decrease.

    A. No, but they are so tiny, they do not interfere with downstream applications.

  • Q. ARE EASYSEP® MAGNETIC NANOPARTICLES FACS-COMPATIBLE?

    A. Yes, the EasySep® particles are flow cytometry-compatible, very uniform in size and ~5000X smaller than other commercially available magnetic beads used with column-free systems.

  • Q. CAN EASYSEP® BE USED TO ISOLATE RARE CELLS?

    A. Yes, however we recommend a cell concentration of 2X108 cells/ml and a minimum working volume of 100 µL. 2x107 cells or fewer should be suspended in 100 µL of buffer.

  • Q. WHICH COLUMNS DO I USE?

    A. None, columns are not used in the EasySep® procedure.

Starting with fresh peripheral blood mononuclear cells, the CD2+ cell content of the enriched fraction typically ranges from 86.3% - 98.0%. The CD2 content was assessed using antibodies against CD3 and CD56.
