The Estrogen Receptor (ER) Alpha protein is a 66 kDa member of the nuclear receptor superfamily of transcription factors. ER Alpha is expressed in the brain, mammary gland, cardiovascular system, urogenital system, uterus and bone. There are 6 structural domains to the receptor, including a DNA binding domain and a ligand binding domain (LBD). Binding of a ligand to the LBD causes a conformational change in the receptor such that it can bind to specific DNA sequences (Estrogen Response Elements or EREs) and can act as a molecular switch to turn on transcriptional activity. Expression of the ER is an important prognostic marker in breast cancer with the presence of ER correlated with better prognosis and the likelihood of response to hormonal therapy.
