EasySep® box
The EasySep® Mouse CD11b Positive Selection Kit is designed to isolate CD11b+ cells from single cell suspensions of splenocytes or other tissues by positive selection. The cocktail contains a combination of PE-labeled antibodies that is directed against CD11b, which are then recognized by Tetrameric Antibody Complexes that are directed against PE and dextran. Labeled cells are bound to magnetic particles and separated using an EasySep® magnet without the use of columns. Cells of interest remain in the tube while unwanted cells are poured off.

  • Q. HOW DOES THE SEPARATION WORK?

    A. Tiny magnetic particles are attached to cells. When placed in the EasySep® Magnet, the cells migrate to the wall of the tube.

  • Q. WHICH COLUMNS DO I USE?

    A. None, columns are not used in the EasySep® procedure.

  • Q. CAN I ALTER THE SEPARATION TIME IN THE MAGNET?

    A. Yes, but this may affect separation performance.

  • Q. IN POSITIVE SELECTION CAN I DO MORE THAN 3 SEPARATIONS TO INCREASE PURITY?

    A. Yes, purity of targeted cells will increase with additional rounds of separations, however the recovery of targeted cells may decrease.

  • Q. ARE EASYSEP® MAGNETIC NANOPARTICLES FACS-COMPATIBLE?

    A. Yes, the EasySep® particles are flow cytometry-compatible, very uniform in size and ~5000X smaller than other commercially available magnetic beads used with column-free systems.

  • Q. CAN THE PARTICLE BE REMOVED AFTER ENRICHMENT?

    A. No, but they are so tiny, they do not interfere with downstream applications.

  • Q. CAN EASYSEP® BE USED TO ISOLATE RARE CELLS?

    A. Yes, however we recommend a cell concentration of 2X108 cells/ml and a minimum working volume of 100 µL. 2x107 cells or fewer should be suspended in 100 µL of buffer.

  • Q. CAN EASYSEP® BE USED FOR EITHER POSITIVE OR NEGATIVE SELECTION?

    A. Yes, we have products for both.

  • Q. HOW CAN I ANALYZE THE PURITY OF MY ENRICHED SAMPLE?

    A. Each Product Information Sheet has detailed staining information.

  • Q. HOW DOES THE BINDING OF THE NANOPARTICLE AFFECT THE FUNCTION OF CELLS? IS THE FUNCTION OF POSITIVELY SELECTED CELLS ALTERED BY THE BOUND PARTICLES?

    A. No, we have found that cells are not functionally altered.
