Primary human mononuclear cells (MNCs) are isolated from umbilical cord blood (CB). Umbilical CB from a normal delivery is collected into a 300 mL standard blood bag containing acid-citrate-dextrose (ACD) anticoagulant (60 mL/bag). Samples are processed within 24 hours of collection. MNCs are obtained by light density separation (1:2 dilution in phosphate buffered saline and centrifugation over Ficoll™). CD56+ NK cells are isolated from MNCs by positive immunomagnetic selection and are cryogenically preserved.
