The 53-6.7 antibody reacts with the α-chain of the CD8 differentiation antigen of all mouse strains tested. CD8α forms heterodimers with the CD8 ß-chain on the cell membrane of most thymocytes and the MHC class I restricted, cytotoxic/suppressor subpopulation of mature T-lymphocytes. CD8α is expressed without CD8ß on subsets of intra-epithelial lymphocytes in the intestine. Maturation of CD8α+ß+ and CD8α+ß- T cells is believed to occur inside and outside the thymus, respectively.
