Cell isolation

Isolate Hematopoietic Progenitor Cells with EasySep
Hematopoietic progenitor cells can be isolated easily and quickly by either depletion of  lineage-specific markers (for human and mouse), or by the positive selection of CD34 (human), or c-kit and Sca-1 (mouse) using STEMCELL’s immunomagnetic, column-free system, EasySep®.  EasySep® can be performed manually, or can be automated with RoboSep®, the fully automated cell separator.  For the rapid de-bulking of lineage-specific markers from whole blood,  STEMCELL also offers a RosetteSep® kit which crosslinks Lin positive cells to red blood cells for depletion.

Isolated cells are highly functional and immediately ready for downstream assays.

Methylcellulose-based colony assay media

Quantitation of hematopoietic progenitors is commonly done with the use of colony assays, where test cells are plated at low cell density into semi-solid medium containing appropriate growth factors. These assay conditions allow the proliferation and differentiation of progenitor cells into mature cell types, which can then be distinguished morphologically.

Several different semi-solid media have been used for hematopoietic colony assays including methylcellulose, agar and collagen. Methylcellulose is now the standard medium for these assays because it permits improved growth of erythroid colonies over other types of semi-solid support systems (e.g. agar), while allowing optimal growth of granulocyte and macrophage colonies. Committed progenitors for both erythroid and granulocyte/macrophage lineages, as well as multi-potential progenitors, can therefore be assayed simultaneously in the same culture dish.

Collagen-based colony assay media

Collagen gels have also been shown to support the proliferation of hematopoietic progenitors. They have the advantage that they can be dehydrated and fixed for subsequent cytochemical and immunocytochemical analysis, making them an ideal system for the detection and quantitation of megakaryocytic progenitors or other progenitors that require more detailed analysis than the standard morphological criteria that are commonly used. Furthermore, since collagen gels can be fixed, long-term records can be established.
 

Long-term culture media

Long-term Culture of Hematopoietic Progenitors
Long-term culture of hematopoietic progenitors is primarily used to quantitate those progenitors capable of initiating and sustaining myelopoiesis for several weeks in vitro. These cells share functional and phenotypic properties with in vivo repopulating cells and are commonly referred to as long-term culture-initiating cells (LTC-IC). MyeloCult® long-term culture media promote the formation of supportive human or mouse stromal layers and allow the proliferation and differentiation of primitive hematopoietic progenitors.

Sera

Serum for Hematopoietic Cell Growth and Differentiation
Fetal bovine sera (FBS) and horse sera are rigorously screened and selected based on their ability to support hematopoietic cell growth and differentiation. Sera are available for several applications, including myeloid colony-forming cell assays, myeloid long-term cultures, pre-B lymphoid colony-forming cell assays and B-lymphoid long-term cultures.

Serum-free media

Serum-free Cell Culture Media for Hematopoietic Cell Growth and Differentiation
Historically, medium containing fetal bovine serum (FBS) has been used to culture hematopoietic cells. However, the concentrations of inhibitors including transforming growth factor-beta and other constituents vary between batches of sera. In order to minimize these variables, particularly for studies of the effects of specific regulatory or growth factors, there has been increased interest in the use of serum-free media. A variety of serum-free media is available for many applications.

ALDEFLUOR – Detect Hematopoietic Stem & Progenitor Cells
The ALDEFLUOR® and ALDECOUNT® reagent systems are non-immunological methods to identify human stem/progenitor cells from bone marrow, peripheral blood, apheresis collections and umbilical cord blood on the basis of their aldehyde dehydrogenase (ALDH) activity. High ALDH activity has also been found in other cell types. Only cells with an intact cellular membrane can retain the ALDEFLUOR®/ALDECOUNT® reaction product, therefore only viable ALDHbr cells are identified. Dead and/or dying cells without intact cellular membranes will not be counted.
